To be blunt, that is a dumb question!

You use it!

If you have created it using the datfile manager then simply select it from the pull down menu and it should start to load it.

Then add a rom path for the dat file and it will search and verify roms turning bad roms RED, perfect roms GREEN and ROMS that are either missing some roms but are available or need renaming to YELLOW.
