It seems RomCenter 3.7.1 is not compatible with the updated XML structure of the newest MAME releases. This is a possible work-around:
1. Download and install Romulus from http://romulus.net63.net/ .
2. Start Romulus and select Add DAT from MAME based EXE.
3. Go to the scanner tab.

Under Options -> General, I am unable to select the backup folder and temp files folder using the selection icons to the right of the text field. I can type into the text field.

So I think I am probably missing something somewhere, which will solve most problems. First question:
